Solid phase extraction (SPE) cleans up and concentrates a sample before HPLC, LC-MS or GC. Three choices decide the cartridge: the sorbent, the sorbent mass and the barrel size.

Step 1: Sorbent: match it to your analyte

SorbentRetentionTypical use
HLB (polymer, 80 Å)Reversed phase for polar and non-polar compoundsGeneral method development, drugs and metabolites in water, plasma or urine; water-wettable, so it tolerates the bed running dry
C18 (silica, 60 Å)Reversed phase, non-polarNon-polar and moderately polar analytes from aqueous samples
C18E (end-capped C18)Reversed phase, fewer silanol interactionsAs C18, with less secondary retention of basic compounds
Silica (60 Å)Normal phase, polarPolar compounds from non-polar organic extracts

All sorbents in this range use 40-60 µm particles.

Step 2: Sorbent mass

The sorbent mass sets the capacity. As a rough guide, a cartridge retains a few percent of its sorbent mass in analyte and matrix, so dirty or large samples need more sorbent. More sorbent also needs more solvent to elute, which dilutes the extract. Masses in this range run from 30 mg for small, clean samples to 20 g for large-scale clean-up.

Step 3: Barrel size

BarrelTypical use
1 mlSmall samples and low elution volumes
3 ml and 6 mlRoutine methods on a vacuum manifold
12, 30 and 60 mlLarge sample volumes or high sorbent masses

The basic method

  • Condition the sorbent with organic solvent, then equilibrate with a solvent like your sample.
  • Load the sample at a slow, steady flow.
  • Wash off interferences with a weak solvent.
  • Elute the analytes with a strong solvent and collect them.

SPE cartridges are sold in packs of 10 to 100 with the price per cartridge shown. Email sales@erohe-lab.com with your analyte and matrix if you need help choosing.

Questions buyers ask

HLB or C18: which SPE sorbent should I start with?

HLB is a good first choice for method development because it retains polar and non-polar compounds and tolerates drying. C18 suits non-polar analytes in aqueous samples.

What is the difference between C18 and C18E?

C18E is end-capped: remaining silanol groups on the silica are blocked, which reduces secondary interactions with basic compounds.

How much sorbent do I need?

As a rough guide, capacity is a few percent of the sorbent mass, so larger or dirtier samples need more sorbent. More sorbent also needs more elution solvent.

Can SPE cartridges be reused?

No. They are single use; reuse risks carry-over and lower recovery.

What equipment do I need to run SPE cartridges?

A vacuum manifold or positive pressure unit, or a syringe for small numbers of samples.
